From the beginning, we took on a challenge that still defines our work today: how to better understand the biology of aging and its role in disease. The decision was to take a long-term approach. We started with fundamental research, built technologies to study aging biology at scale, and created an environment where integrated teams could follow the science where it leads us. Today, that foundation supports our progress from discovery into clinical- and early-stage programs across multiple age-related diseases. We recently joined the PKD Foundation community at two of their annual events: PKDCon, which brought together people living with ADPKD, their caregivers, and family members; and the PKD Centers of Excellence, a conference for nephrologists and investigators focused on advancing standards of care. We appreciated hearing from people living with ADPKD, connecting with clinicians, and sharing more about our research investigating PAPP-A. PAPP-A is a protein that influences insulin-like growth factor (IGF) signaling, which may play a role in kidney cyst development and growth in ADPKD. This potential role has not been established and remains under investigation. Genomic deep learning models have significant potential to advance how scientists study DNA and RNA, but computing demands have kept many of these tools out of reach for researchers who could benefit from them. A recent study from Calico’s David Kelley and Han Yuan offers a new framework designed to make these models more accessible. Published in Genome Biology, the Kelley Lab developed and evaluated a transfer learning approach for Borzoi, our model for predicting gene expression profiles from DNA sequence data. As David explains, “For anyone who wants to apply these models to their own data, everything is there. Recently published in Nature, Calico scientists collaborated on new NIH research that advances understanding of immune regulation and how regulatory cells help control tissue inflammation. Co-led by Calico's Chief Scientific Officer Michael Lenardo, M.D., the study was first- authored by Jing Cui, Ph.D., with contributions from Lixin Zheng, M.D., Ph.D. Mike joined Calico from the NIH (where he served as Co-Director of the Clinical Genomics Program at NIAID), bringing deep expertise in immunology and genetics to advancing our mission. We’re pleased to announce the FDA has granted Breakthrough Therapy Designation to fosigotifator, Calico’s investigational treatment for Vanishing White Matter (VWM) disease. VWM is an ultra-rare, progressive disease of the brain's white matter caused by variations in eIF2B, an enzyme essential for protein synthesis. There are no approved treatments. Fosigotifator is designed to target eIF2B directly with the goal of restoring its activity and reducing chronic activation of the Integrated Stress Response in the brain. This designation reflects preliminary clinical data suggesting fosigotifator may have the potential to demonstrate substantial improvement over existing therapies on one or more clinically significant endpoints. For patients and families affected by VWM, Breakthrough Therapy Designation means the FDA will work with Calico to expedite the development and review of fosigotifator. Fosigotifator was previously selected for the FDA’s START program, an initiative focused on accelerating the development of novel treatments for rare diseases. Fosigotifator is an investigational drug. Its safety and efficacy have not been established and it has not been approved by any regulatory authority. Calico’s Head of Clinical Science Bill Cho, M.D., Ph.D., will share updates on Calico’s ongoing research and clinical development efforts in Vanishing White Matter disease, an ultra-rare genetic condition. Cathleen (Cathy) Hartman is the Project Team Leader for our PTPN2 immuno-oncology program. Cathy drives the development of our PTPN2 inhibitor in collaboration with her functional leads and clinical investigators. New opportunities in aging research are increasingly emerging at the intersection of biology and machine learning. Calico’s Head of AI/ML, Matt Onsum, recently took part in the Caltech Longevity Hackathon, where students, researchers, engineers, and founders came together to explore new approaches to aging and longevity science. Throughout the weekend, teams developed technologies spanning biology, neuroscience, AI, and medicine. Matt spoke about how machine learning is advancing drug discovery at Calico and creating new opportunities for scientific progress. We enjoyed the opportunity to connect with a community so deeply engaged in exploring how computation and biology can shape the future of aging research.
